Buthionine Sulfoximine, Glutathione Synthase Inhibitor, Potentiate the Paraquat Toxicity in Mice

Abstract
In biological systems, the repetition of one electron reduction of paraquat results in the continuous formation of radical species which react with molecular oxygen. Antioxidant systems, such as superoxide dismutase (SOD), glutathione peroxidase, catalase, NADH, NADPH, glutathione, vitamin C & E, uric acid, carotene and the like, are involved in the defense of paraquat toxicity. To investigate the effects of glutathione-depletion in paraquat toxicity, total glutathione was diminished by intraperitoneal injection of buthionine sulfoximine (600 mg/kg) in mice weighing about 25 gram, and after 18 hours, paraquat (40 mg/kg) was injected intraperitoneally. 24 Hours later, antioxidant enzymes were more attenuated in the group of combined treatment than paraquat-treatment alone. As a result of these facts we came to a conclusion that the depletion of glutathione causes the aggravation of paraquat toxicity and glutathione is probably one of the major components in the defense of paraquat toxicity.
